Statistical analysis for HDX data is continuously evolving. Crook et. al., in 2022 (Commun Biol 5, 588 (2022)) used a functional data analysis approach to fit HDX uptake curves in order to test their significance. This approach does not test individual HDX differences but uses the whole peptide to do it.

Although this approach is not widely used still, we used the R code developed by Crook et al., and implemented it into our app. Please read their manuscript and familiarize yourself with the code behind it and the results you can expect from it.
